ah... the old "multi-system" sync question :-)
Simply install Plaxo on both your laptop and desktop systems. Both installations of Plaxo should be 'attached' to the same Plaxo account. The registration wizard will ask you if you are a new or existing user. So for the first install, select new (assuming you don't already have a Plaxo account). And for the second, select existing. And that's it. Plaxo will do the rest for you automatically.
One way to think of multi-system sync is this: Your Plaxo account is like the hub of a wheel. And your local machines are like the end-points outside of the hub. The toolbar is the spokes that connect the endpoints to the hub syncing data between each end-point and your Plaxo account. Data that enters into your Plaxo account from one of the end-points can then be synchronized out to all of the other end-points automatically. You can then access your information through one of the end-points, or through the hub (your Plaxo account by logging into Plaxo Online).
As an aside, each end-point can control what information it sync with your Plaxo account, but that's a more advanced topic (see sync manager), and you asked to be gentle :-)
